DAB2 Antibody (YA4142) is a mouse-derived and non-conjugated IgG1 antibody, targeting to DOC2. It can be applicated for ICC/IF, FC, ELISA assays, in the background of human.

DAB2 is an adapter protein functioning as a clathrin-associated sorting protein (CLASP) essential for clathrin-mediated endocytosis of specific cargo proteins. It binds and assembles clathrin while simultaneously interacting with phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2) and cargo proteins containing non-phosphorylated NPXY internalization motifs (e.g., LDL receptor), recruiting them to clathrin-coated pits. DAB2 operates independently of the AP-2 complex in clathrin-mediated endocytosis and participates in integrin β-1 endocytosis (a redundant function with AP-2 requiring DAB2’s binding to EH domain-containing proteins like EPS15, EPS15L1, and ITSN1). It also mediates endocytosis of cystic fibrosis transmembrane conductance regulator (CFTR) and megalin/LRP2 lipoprotein receptor during embryogenesis, as well as TGF-β receptor recycling. DAB2 facilitates CFTR trafficking to late endosomes and engages in multiple receptor signaling pathways, including TGF-β receptor signaling (promoting SMAD2 phosphorylation) and TGF-β-stimulated JNK activation. It may inhibit canonical Wnt/β-catenin signaling by stabilizing the β-catenin destruction complex (via competitive axin binding to block PP1-mediated dephosphorylation) or sequestering LRP6 via clathrin endocytosis, while potentially activating non-canonical Wnt pathways. In growth factor/Ras signaling, DAB2 suppresses ERK activation by disrupting GRB2-SOS1 interaction and inhibits SRC by preventing its phosphorylation at Tyr-419. It may modulate androgen receptor (AR) signaling by competing with AR for SRC binding and contributes to CSF-1 signal transduction, cellular differentiation, visceral endoderm (VE) formation during embryogenesis (responding to epiblast-derived Nodal signals), and epithelial-mesenchymal transition (EMT). Additionally, DAB2 might regulate myeloid differentiation, induce macrophage adhesion/spreading, and act as a tumor suppressor.
