Background

DNA PKcs is a serine/threonine-protein kinase that functions as a molecular sensor for DNA damage. It plays a crucial role in DNA non-homologous end joining (NHEJ), which is essential for double-strand break (DSB) repair and V(D)J recombination. DNA PKcs must bind to DNA to exhibit its catalytic activity and promotes the processing of hairpin DNA structures in V(D)J recombination by activating the hairpin endonuclease artemis (DCLRE1C). Recruited by XRCC5 and XRCC6 to DNA ends, it protects and aligns broken DNA ends, prevents their degradation, and sequesters DSBs for NHEJ-mediated repair. Additionally, DNA PKcs acts as a scaffold protein to facilitate the localization of DNA repair proteins to damage sites, and the assembly of the DNA-PK complex at DNA ends is required for the NHEJ ligation step. It is also found at chromosome ends, suggesting a potential role in maintaining telomeric stability and preventing chromosomal end fusion (by similar). DNA PKcs is involved in transcriptional modulation and, as part of the DNA-PK complex, contributes to early ribosome assembly by promoting the processing of precursor rRNA into mature 18S rRNA. It recognizes the substrate consensus sequence [ST]-Q and phosphorylates histone variant H2AX at Ser-139, thereby regulating the DNA damage response mechanism. Furthermore, DNA PKcs phosphorylates various proteins, including ASF1A, DCLRE1C, c-Abl/ABL1, histone H1, HSPCA, c-jun/JUN, p53/TP53, PARP1, and others. It can phosphorylate C1D in the presence of supercoiled DNA, and its ability to phosphorylate p53/TP53 under such conditions depends on C1D. DNA PKcs regulates the phosphatidylinositol 3-kinase/protein kinase B signaling pathway by mediating phosphorylation of PKB/AKT1, PKB/AKT2, and PKB/AKT3 at Ser-473, promoting their activation. It also modulates circadian period length by antagonizing CRY1 Ser-588 phosphorylation and increasing CRY1 protein stability (by similar). Additionally, DNA PKcs plays a role in regulating DNA virus-mediated innate immune responses by assembling into the HDP-RNP complex, which serves as a platform for IRF3 phosphorylation and subsequent activation of the innate immune response via the cGAS-STING pathway. It further regulates this pathway by phosphorylating CGAS, impairing its oligomerization and activation, and by mediating PARP1 phosphorylation.
