The DKK-3 protein functions as an antagonist of canonical Wnt signaling by impeding the interaction between LRP5/6 and Wnt and forming a ternary complex with the transmembrane protein KREMEN, facilitating the internalization of LRP5/6. Within vertebrate development, DKKs, including DKK-3, play a crucial role by locally inhibiting Wnt-regulated processes such as antero-posterior axial patterning, limb development, somitogenesis, and eye formation. In the adult, DKKs are implicated in diverse physiological and pathological conditions, encompassing bone formation, bone disease, cancer, and Alzheimer's disease (By similarity).
