DDX3 Antibody (YA5296) is a Mouse-derived and non-conjugated monoclonal antibody, targeting to DDX3.

DDX3 is a multifunctional ATP-dependent RNA helicase whose ATPase activity can be stimulated by various ribo- and deoxyribonucleic acids, indicating relaxed substrate specificity. It can unwind partially double-stranded DNA with a preference for 5'-single-stranded DNA overhangs and binds RNA G-quadruplex (rG4s) structures. DDX3 is involved in numerous cellular processes, including transcription regulation, translation initiation, and innate immunity. In transcription regulation, it positively regulates CDKN1A/WAF1/CIP1 transcription in an SP1-dependent manner to inhibit cell growth while repressing CDH1/E-cadherin transcription. In translation regulation, DDX3 promotes efficient translation of selected mRNAs with complex 5'-UTRs and facilitates the assembly of functional 80S ribosomes independently of its ATPase activity. In innate immunity, DDX3 acts as a viral RNA sensor, binding viral RNAs and promoting type I interferon (IFN-alpha and IFN-beta) production. Additionally, DDX3 participates in stress granule assembly, NLRP3 inflammasome regulation, and activation of the Wnt/beta-catenin signaling pathway. During viral infections, DDX3 facilitates the replication of HCV, HIV-1, ZIKV, DENV, and VEEV.
