DDX21 is an RNA helicase that senses the transcriptional status of both RNA polymerase (Pol) I and II, promoting ribosomal RNA (rRNA) processing and Pol II transcription. It binds various RNAs, including rRNAs, snoRNAs, 7SK RNA, and, to a lesser extent, mRNAs. In the nucleolus, DDX21 localizes to the rDNA locus, directly binding rRNAs and snoRNAs to facilitate rRNA transcription, processing, and modification. It is required for rRNA 2'-O-methylation, likely by recruiting late-acting snoRNAs SNORD56 and SNORD58 to pre-ribosomal complexes. In the nucleoplasm, DDX21 binds 7SK RNA and is recruited to Pol II-transcribed gene promoters, where it promotes transcription by releasing P-TEFb from inhibitory 7SK snRNP in a helicase-dependent manner. Additionally, DDX21 acts as a cofactor for JUN-activated transcription, facilitating phosphorylation of JUN at Ser-77. It exhibits helicase activity (unwinding double-stranded RNA) and foldase activity (folding single-stranded RNA). In collaboration with SIRT7, DDX21 prevents R-loop-associated DNA damage and transcription-related genomic instability by overcoming R-loop-mediated RNA polymerase stalling through SIRT7-mediated deacetylation. DDX21 is also involved in rRNA processing and may bind specific miRNA hairpins. By similarity, DDX21 is part of a multi-helicase-TICAM1 complex that serves as a cytoplasmic sensor of viral double-stranded RNA (dsRNA), triggering antiviral responses, including pro-inflammatory cytokine induction, via the adapter TICAM1.
