Background

DDX17 is an RNA helicase that unwinds RNA and alters RNA structures through ATP binding and hydrolysis. It is involved in multiple cellular processes, including pre-mRNA splicing, alternative splicing, ribosomal RNA processing, miRNA processing, and transcription regulation. DDX17 regulates the alternative splicing of exons with specific features, such as promoting the inclusion of AC-rich alternative exons in CD44 transcripts, a function dependent on its RNA helicase activity. It also affects NFAT5 and histone macro-H2A.1/MACROH2A1 alternative splicing in a CDK9-dependent manner. In NFAT5, DDX17 promotes the inclusion of exon 4 containing stop codons, potentially leading to NFAT5 protein downregulation. Additionally, DDX17 regulates splicing of mediators in the steroid hormone signaling pathway, including kinases (e.g., CDK2, MAPK1, GSK3B) and transcriptional regulators (e.g., CREBBP, MED1, NCOR1, NCOR2). Collaborating with HNRNPH1, it controls splicing of specific exon subsets in myoblasts and epithelial cells. Beyond mature mRNAs, DDX17 interacts with pri-microRNAs (e.g., MIR663/miR-663a, MIR99B/miR-99b, MIR6087/miR-6087) via a 5'-[ACG]CAUC[ACU]-3' consensus sequence. It is essential for the production of microRNAs like MIR21 and MIR125B1 and may stabilize pri-microRNAs (By similarity). At high cell density, DDX17 contributes to MYC downregulation by generating MYC-targeting microRNAs. Alongside DDX5, it may process the 32S intermediate into mature 28S ribosomal RNA. As a promoter-specific transcriptional regulator, DDX17 acts as a coactivator or corepressor depending on context. It enhances NFAT5 transcriptional activity and synergizes with TP53 to activate the MDM2 promoter (requiring lysine acetylation). DDX17 may also coactivate MDM2 independently of TP53 and coactivates MMP7 transcription. With CTNNB1, it coactivates MYC, JUN, FOSL1, and CCND1 transcription. Alone or with DDX5/SRA1 non-coding RNA, DDX17 promotes assembly of transcription initiation complexes and chromatin remodeling for MYOD1-dependent transcription, a helicase-independent activity critical for skeletal muscle differentiation. During epithelial-to-mesenchymal transition, DDX17 coregulates SMAD-dependent transcription, directly controlling differentiation effectors, including miRNAs that repress its expression. In estrogen and testosterone signaling, DDX17 mediates alternative promoter usage and regulates transcription/splicing of steroid hormone target genes. Unlike splicing regulation, its ESR1 transcriptional coregulation is helicase-independent. In innate immunity, DDX17 restricts bunyavirus infection (e.g., RVFV, LACV) independently of interferon and DROSHA, binding RVFV RNA via structured viral elements and promoting ZC3HAV1-mediated mRNA degradation in an ATPase-dependent manner.
