DBC1 Antibody (YA5182) is a mouse-derived and non-conjugated monoclonal antibody, targeting to DBC1. It can be applicated for WB, ICC/IF, IP assays, in the background of human, mouse, rat, monkey.

Background

DBC 1 is a core component of the DBIRD complex, a multiprotein complex that operates at the interface between core mRNP particles and RNA polymerase II (RNAPII), integrating transcript elongation with the regulation of alternative splicing. The DBIRD complex modulates local elongation rates and alternative splicing of exons embedded in (A + T)-rich DNA regions. It inhibits SIRT1 deacetylase activity, leading to increased p53/TP53 acetylation and p53-mediated apoptosis. Additionally, DBC 1 suppresses SUV39H1 methyltransferase activity, mediates ligand-dependent transcriptional activation by nuclear hormone receptors, and plays a critical role in maintaining genomic stability after UV-induced genotoxic stress. It regulates the circadian expression of core clock components NR1D1 and BMAL1 and enhances NR1D1's transcriptional repressor activity by stabilizing its protein levels. DBC 1 negatively regulates HDAC3 deacetylase activity and may alter its subcellular localization while positively influencing the β-catenin pathway (canonical Wnt signaling). It also contributes to tumor suppression through p53/TP53 regulation by stabilizing p53/TP53 via interference with its interaction with the ubiquitin ligase MDM2.
