Description

Daxx Antibody (YA4810) is a mouse-derived and non-conjugated monoclonal antibody, targeting to Daxx. It can be applicated for WB, ICC/IF, FC, ELISA assays, in the background of human.

Background

Daxx is a transcription corepressor that represses the transcriptional potential of several sumoylated transcription factors, downregulating both basal and activated transcription. Its repressor activity is modulated by recruitment to subnuclear compartments like the nucleolus or PML/POD/ND10 nuclear bodies via interactions with MCSR1 and PML, respectively. It cooperates with PML in PML/POD/ND10 bodies to regulate transcription, potentially influencing TNFRSF6-dependent apoptosis. Daxx inhibits transcriptional activation of PAX3 and ETS1 through direct protein interactions and modulates PAX5 activity (possibly involving CREBBP). As an adapter in the MDM2-DAXX-USP7 complex, it regulates MDM2 ubiquitination: under non-stress conditions, DAXX-USP7 binding prevents MDM2 autoubiquitination, enhancing MDM2-mediated TP53 ubiquitination and degradation; DNA damage disrupts the complex, increasing MDM2 autoubiquitination and stabilizing TP53. Daxx acts as a histone chaperone for H3.3 deposition and, as part of the ATRX:DAXX remodeling complex, catalyzes replication-independent H3.3 incorporation at pericentric repeats and telomeres. During neuronal activation, it deposits H3.3 at immediate early gene regulatory elements, potentially linked to their induction. DAXX also recruits H3.3:H4 dimers to PML-NBs (ATRX-independently but facilitated by ASF1A) and mediates mislocalization of overexpressed CENPA to chromosomes. It activates the JNK pathway and apoptosis via MAP3K5 in response to TNFRSF6/TGFBR2 signaling, though HSPB1/HSP27 binding can block this. Notably, lymphoid cell JNK activation and TNFRSF6-mediated apoptosis may be DAXX-independent. Daxx exhibits restriction activity against HCMV and positively regulates HSF1 during the stress response.
