Cytochrome P450 2C9 is a cytochrome P450 monooxygenase involved in the metabolism of various endogenous substrates, including fatty acids and steroids. Mechanistically, it uses molecular oxygen to insert one oxygen atom into a substrate while reducing the second into a water molecule, with electrons provided by NADPH via cytochrome P450 reductase (NADPH--hemoprotein reductase). This enzyme catalyzes the epoxidation of polyunsaturated fatty acid (PUFA) double bonds and the hydroxylation of carbon-hydrogen bonds. In cholesterol metabolism, it converts cholesterol to 25-hydroxycholesterol, a physiological regulator of cellular cholesterol homeostasis. It also exhibits low catalytic activity in forming catechol estrogens from 17β-estradiol (E2) and estrone (E1), such as 2-hydroxy E1 and E2. Additionally, it catalyzes bisallylic hydroxylation and hydroxylation with double-bond migration in PUFAs. Similar functions include metabolizing plant monoterpenes like limonene, oxygenating it to produce carveol and perillyl alcohol. Cytochrome P450 2C9 also contributes to the pharmacokinetic variability of drugs such as S-warfarin, diclofenac, phenytoin, tolbutamide, and losartan.

  Western blot analysis was performed on extracts from HepG2 (lane 1, 20 μg), using CYP2C9 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Mouse/Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
