Cullin 5 is a core component of multiple cullin-5-RING E3 ubiquitin-protein ligase complexes (ECS complexes, also named CRL5 complexes), which mediate ubiquitination and subsequent proteasomal degradation of target proteins. It acts as a scaffold protein that contributes to catalysis by positioning the substrate and the ubiquitin-conjugating enzyme. The functional specificity of the E3 ubiquitin-protein ligase complex depends on the variable SOCS box-containing substrate recognition component. During cortex development, Cullin 5 acts as a key regulator of neuron positioning, being part of various SOCS-containing ECS complexes (e.g., ECS(SOCS7)) that regulate reelin signaling by mediating ubiquitination and degradation of DAB1 (by similar). ECS(SOCS1) may direct ubiquitination of JAK2, while ECS(SOCS2) mediates ubiquitination and degradation of phosphorylated EPOR and GHR. ECS(SPSB3) catalyzes ubiquitination of nuclear CGAS. The ECS(KLHDC1) complex is part of the DesCEND pathway and mediates ubiquitination and degradation of truncated SELENOS selenoprotein produced by failed UGA/Sec decoding. ECS(ASB9) mediates ubiquitination and degradation of CKB. Some ECS complexes promote 'Lys-11'-linked ubiquitination and degradation of BTRC, whereas a multisubunit ECS complex polyubiquitinates monoubiquitinated POLR2A. ECS(RAB40C) mediates ANKRD28 ubiquitination and degradation, inhibiting protein phosphatase 6 (PP6) complex activity and focal adhesion assembly during cell migration. ECS(RAB40A) mediates 'Lys-48'-linked ubiquitination and degradation of RHOU, inhibiting focal adhesion disassembly. ECS(RAB40B) mediates ubiquitination of LIMA1/EPLIN and RAP2, regulating actin cytoskeleton dynamics and stress fiber formation during cell migration. Cullin 5 may also form a cell surface vasopressin receptor. In microbial infection, following HIV-1 infection, CUL5 associates with HIV-1 Vif proteins to form a cullin-5-RING E3 ubiquitin-protein ligase complex that ubiquitinates and degrades APOBEC3F and APOBEC3G, with partial ubiquitination of APOBEC3H. Additionally, Cullin 5 appears to be involved in proteasomal degradation of p53/TP53 stimulated by adenovirus E1B-55 kDa protein.
