Cullin 3 Antibody (YA2578) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Cullin 3.

Background

Cullin 3 is a core component of multiple cullin-RING-based BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complexes, which mediate the ubiquitination and subsequent proteasomal degradation of target proteins. The BCR complexes collaborate with ARIH1 to ubiquitinate target proteins. As a scaffold protein, Cullin 3 may contribute to catalysis by positioning the substrate and ubiquitin-conjugating enzyme. The E3 ubiquitin-protein ligase activity of the complex depends on the neddylation of the cullin subunit and is inhibited by the association of the deneddylated cullin subunit with TIP120A/CAND1. The functional specificity of the BCR complex is determined by the BTB domain-containing protein as the substrate recognition component. For example, BCR(KLHL42) is involved in the ubiquitination of KATNA1, while BCR(SPOP) targets BMI1/PCGF4, BRMS1, MACROH2A1, DAXX, GLI2, and GLI3. Cullin 3 can also form BCR complexes containing homodimeric SPOPL or the SPOP-SPOPL heterodimer, which exhibit lower ubiquitin ligase activity. Additionally, BCR(KLHL9-KLHL13) regulates AURKB dynamics on mitotic chromosomes, ensuring faithful mitotic progression and cytokinesis completion. BCR(KLHL12) modulates ER-Golgi transport by controlling COPII coat size, critical for collagen export. BCR(KLHL3) regulates ion transport in the distal nephron. BCR(KLHL20) participates in interferon response and anterograde Golgi-to-endosome transport. BCR(KLHL21) controls the chromosomal passenger complex (CPC) localization. BCR(KLHL22) mediates PLK1 monoubiquitination. BCR(KLHL25) maintains translational homeostasis and lipid synthesis. BCR(KBTBD8) remodels the translational program for neural crest specification. BCR(KCTD17) promotes ciliogenesis by degrading TCHP. BCR(KLHL24) regulates KRT14 levels during keratinocyte differentiation. BCR(KLHL18) activates AURKA at the centrosome. BCR(KEAP1) senses oxidative stress by degrading NFE2L2/NRF2. CUL3(KBTBD6/7) ubiquitinates TIAM1, regulating RAC1 signaling. BCR(KBTBD4) targets CoREST complex components for degradation. BCR(ARMC5) mediates premature transcription termination by ubiquitinating POLR2A. Cullin 3 also facilitates 'Lys-63'-linked ubiquitination of MRPL12.
