Background

Cullin 1 is a core component of multiple cullin-RING-based SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes, which mediate the ubiquitination of proteins involved in cell cycle progression, signal transduction, and transcription. The SCF complexes collaborate with ARIH1 to ubiquitinate target proteins. Within the SCF complex, Cullin 1 acts as a rigid scaffold organizing SKP1-F-box protein and RBX1 subunits, potentially contributing to catalysis by positioning the substrate and ubiquitin-conjugating enzyme. The E3 ubiquitin-protein ligase activity of the complex depends on cullin subunit neddylation, and substrate recognition component exchange is mediated by TIP120A/CAND1. The functional specificity of SCF complexes is determined by the F-box protein. For example, SCF(BTRC) and SCF(FBXW11) ubiquitinate CTNNB1 and participate in Wnt signaling; SCF(SKP2) ubiquitinates phosphorylated CDKN1B/p27kip, regulating G1/S transition; SCF(FBXW7) ubiquitinates CCNE1 and the NOTCH1 intracellular domain (NICD). Additionally, SCF complexes ubiquitinate various other proteins, including NFKBIA, NFKBIB, CDC25A, and CEP68. by similar: The SCF(FBXO31) complex specifically ubiquitinates C-terminally amidated proteins in response to oxidative stress.
