Background

CUG BP1 is an RNA-binding protein involved in regulating various post-transcriptional events, including pre-mRNA alternative splicing, mRNA translation, and stability. It mediates exon inclusion and/or exclusion in tissue-specific and developmentally regulated alternative splicing, such as specifically activating exon 5 inclusion in cardiac isoforms of TNNT2 during heart remodeling. The protein acts as both an activator and repressor of coregulated exons: promoting the inclusion of the smooth muscle (SM) exon while excluding the non-muscle (NM) exon in actinin pre-mRNAs. It activates SM exon 5 inclusion by antagonizing PTB's repressive effect and promotes the exclusion of exon 11 in INSR pre-mRNA. Together with HNRNPH1, it inhibits insulin receptor (IR) pre-mRNA exon 11 inclusion in myoblasts. Additionally, CUG BP1 regulates the translation initiation codon selection of CEBPB mRNA and enhances its translation in aging liver (By similarity). It increases CDKN1A mRNA translation by counteracting CALR3's repressive effect and mediates rapid cytoplasmic mRNA deadenylation by recruiting the PARN deadenylase to EDEN-containing mRNAs. The protein is required for spermatogenesis completion (By similarity). CUG BP1 binds to diverse RNA sequences, including (CUG)n triplet repeats in the 3'-UTR of DMPK, Bruno response elements (BREs), muscle-specific splicing enhancer (MSE) sites flanking TNNT2 exon 5, AU-rich sequences (AREs or EDEN-like) in the 3'-UTR of JUN and FOS mRNAs, IR RNA, and the 5'-regions of CDKN1A and CEBPB mRNAs. It may also act as a specific regulator of miRNA biogenesis by binding to primary microRNA pri-MIR140 and, along with CELF2, negatively regulating its processing into mature miRNA.
