CRTC1 is a transcriptional coactivator that activates transcription by interacting with CREB1, acting on both consensus and variant cAMP response element (CRE) sites. Within the SIK/TORC signaling pathway, CRTC1 functions as a coactivator, with its activity dependent on dephosphorylation and independent of CREB1 Ser-133 phosphorylation. It enhances the interaction between CREB1 and TAF4 and regulates the expression of specific CREB-activated genes, such as the steroidogenic gene StAR. CRTC1 is also a potent coactivator of PGC1alpha, inducing mitochondrial biogenesis in muscle cells. In the hippocampus, CRTC1 is involved in the maintenance of late-phase long-term potentiation (L-LTP) at Schaffer collateral-CA1 synapses. Additionally, CRTC1 may be essential for dendritic growth in developing cortical neurons (by similarity). CRTC1 collaborates with SIK1 to regulate light-induced entrainment of the circadian clock. Upon light stimulation, CRTC1 coactivates CREB-mediated transcription of PER1, which plays a critical role in the photic entrainment of the circadian clock. Furthermore, during microbial infection, CRTC1 acts as a coactivator for TAX-mediated activation of the human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR).
