CREB Regulated Transcription Coactivator 2 (CRTC2) is a transcriptional coactivator for CREB1, activating transcription through both consensus and variant cAMP response element (CRE) sites. It functions as a coactivator in the SIK/TORC signaling pathway, becoming active upon dephosphorylation and acting independently of CREB1 'Ser-133' phosphorylation. CRTC2 enhances the interaction between CREB1 and TAF4. As part of the LKB1/AMPK/TORC2 signaling pathway, it regulates gluconeogenesis and controls the expression of specific genes, such as the steroidogenic gene StAR. CRTC2 is also a potent coactivator of PPARGC1A, promoting mitochondrial biogenesis in muscle cells. Additionally, it serves as a coactivator for TAX-mediated activation of the human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR).
