CPT1A catalyzes the transfer of the acyl group from long-chain fatty acid-CoA conjugates to carnitine, a critical step for mitochondrial uptake of long-chain fatty acids and their subsequent beta-oxidation in mitochondria. Additionally, CPT1A exhibits lysine succinyltransferase activity, regulating the enzymatic activity of substrate proteins (e.g., ENO1) and metabolism independently of its classical carnitine O-palmitoyltransferase function. It plays a significant role in hepatic triglyceride metabolism (by similar). Upon activation by butyryl-CoA, CPT1A contributes to inducible regulatory T-cell (iTreg) differentiation by counteracting malonyl-CoA-mediated repression (by similar). It sustains the IFN-I response by recruiting ZDHHC4 to palmitoylate MAVS at mitochondria, stabilizing and activating MAVS. Furthermore, CPT1A promotes ROS-induced oxidative stress in liver injury via modulation of NFE2L2 and NLRP3-mediated signaling pathways (by similar).
