COX2 Antibody (YA5631) is a mouse-derived and non-conjugated IgG2b monoclonal antibody, targeting to COX2. It can be applicated for IHC-P, WB, ICC/IF, ELISA assays, in the background of human.

COX2 is a bifunctional enzyme with both cyclooxygenase and peroxidase activities, involved in the biosynthesis pathway of prostanoids, primarily derived from arachidonate (AA) and playing a key role in inflammatory responses. Its cyclooxygenase activity oxygenates AA to prostaglandin G2 (PGG2), which is then reduced by peroxidase activity to prostaglandin H2 (PGH2), the precursor of all 2-series prostaglandins and thromboxanes. Similarly, COX2 catalyzes the conversion of dihomo-gamma-linoleate (DGLA) and eicosapentaenoate (EPA) to PGH1 and PGH3, precursors of 1-series and 3-series prostaglandins, respectively. Additionally, COX2 metabolizes 2-arachidonoyl lysophospholipids and 2-arachidonoyl glycerol, contributing to prostanoid lysophospholipid production and pain response. The enzyme also generates lipid mediators from n-3 and n-6 polyunsaturated fatty acids (PUFAs) via a lipoxygenase-type mechanism and produces resolvins during inflammation resolution, such as 17R-HDHA (a precursor of D-series resolvins) from docosahexaenoate (DHA) and 18S-HEPE (a precursor of E-series resolvins) from eicosapentaenoate (EPA). In vascular endothelial cells, COX2 converts docosapentaenoate (DPA) to 13R-HDPA, a precursor of 13-series resolvins (RvTs) that activate macrophage phagocytosis. In activated leukocytes, it oxygenates hydroxyeicosatetraenoates (HETEs) to diHETEs. COX2 can also utilize linoleate (LA) as a substrate to produce (9R)-HODE and (13S)-HODE in a regio- and stereospecific manner (By similarity). During neuroinflammation, COX2 is involved in neuronal secretion of 15R-lipoxin A4, which regulates phagocytic microglia (By similarity).
