Complement factor B (CFB) is the precursor of the catalytic component of the C3 and C5 convertase complexes in the alternative pathway of the complement system, a protein cascade that facilitates pathogen phagocytosis, degradation, and adaptive immune signaling enhancement. The alternative pathway acts as an amplification loop by promoting additional C3 and C5 convertase formation to augment other complement pathways (classical, lectin, and GZMK). Upon cleavage and activation by factor D (CFD), CFB generates Ba and Bb chains, with the Bb chain serving as the catalytic subunit of C3/C5 convertases. As a serine protease component, CFB forms the C3 convertase with complement C3b, cleaving C3 into C3a anaphylatoxin and C3b opsonin. Binding of an additional C3b molecule to the C3 convertase forms the C5 convertase, which activates C5 into C5a anaphylatoxin and the membrane attack complex component C5b.
