Clusterin alpha Chain Antibody (YA2525) can be used for WB, IP experiment in rat background.

Background

Clusterin alpha Chain functions as an extracellular chaperone that prevents aggregation of non-native proteins, including stress-induced aggregation of plasma proteins (By similarity). It inhibits amyloid fibril formation by APP, APOC2, B2M, CALCA, CSN3, SNCA, and aggregation-prone LYZ variants in vitro (PubMed:14741101). ATP-independent, it maintains partially unfolded proteins for refolding by other chaperones like HSPA8/HSC70 but lacks autonomous refolding activity. Binding to cell surface receptors triggers internalization and lysosomal/proteasomal degradation of client complexes. Secreted forms protect against apoptosis and complement-mediated cytolysis by blocking C9 polymerization in the MAC (By similarity). Intracellularly, it interacts with ubiquitin and SCF E3 ligases to promote degradation of targets (e.g., COMMD1, IKBKB) and modulates NF-kappa-B activity (By similarity). Under stress, it can either promote apoptosis (PubMed:12551933) or inhibit it via mitochondrial BAX-cytochrome c interference. It regulates proliferation and, during ER stress, stabilizes mitochondrial membranes via HSPA5 binding (By similarity). Secreted forms do not affect caspase/BAX-mediated apoptosis or TNF-induced NF-kappa-B (By similarity) but modulate neuronal differentiation through STMN3 interaction (By similarity) and clear injury-related immune complexes (PubMed:11865066).
