CLEC12A Antibody (YA4328) is a mouse-derived and non-conjugated IgG2a antibody, targeting to CLL1. It can be applicated for WB, ICC/IF, FC, ELISA assays, in the background of human.

CLEC12A is a myeloid inhibitory C-type lectin receptor that acts as a negative regulator of myeloid cell activation. It recognizes and binds various structures, such as neutrophil extracellular traps (NETs) or monosodium urate crystals, to inhibit excessive myeloid cell activation and limit proinflammatory pathways (by similar). Additionally, CLEC12A serves as a pattern-recognition receptor for pathogen-associated molecules, including plasmodium hemozoin or mycobacterial micolic acid. Ligand binding induces phosphorylation of its ITIM motif, leading to recruitment of tyrosine-protein phosphatases PTPN6 and PTPN11, which counteract tyrosine-protein kinase SYK to prevent myeloid cell activation. In neutrophils, CLEC12A specifically recognizes DNA in NETs to inhibit neutrophil activation and limit further NET formation (by similar). It also acts as a receptor for monosodium urate crystals to down-regulate neutrophil activation and promote the type I interferon response (by similar). Furthermore, CLEC12A inhibits natural killer (NK) cell cytotoxicity and suppresses dendritic cell maturation in an IL10-dependent manner.
