Claudin 1 (CLDN1) is a major component of tight junction complexes that regulate epithelial permeability. Coexpressed and interacting with other claudin family members, it determines overall barrier properties. CLDN1 prevents paracellular diffusion of small molecules in the epidermis and is essential for normal skin barrier function. It maintains water homeostasis indirectly by modulating expression of other proteins (though dispensable for water barrier formation in keratinocyte tight junctions). Additionally, CLDN1 serves as a co-receptor for hepatitis C virus (HCV) by forming the CLDN1-CD81 complex critical for viral entry, and also acts as a receptor for dengue virus.
