cIAP2 is a multi-functional protein that regulates caspases and apoptosis, as well as modulates inflammatory signaling and immunity, mitogenic kinase signaling and cell proliferation, and cell invasion and metastasis. It acts as an E3 ubiquitin-protein ligase to regulate NF-kappa-B signaling, exerting opposing effects on canonical and non-canonical NF-kappa-B pathways: promoting the canonical pathway while suppressing constitutive activation of the non-canonical pathway. Its E3 ubiquitin-protein ligase targets include RIPK1, RIPK2, RIPK3, RIPK4, CASP3, CASP7, CASP8, IKBKE, TRAF1, and BCL10. Additionally, cIAP2 serves as a key regulator of innate immune signaling by modulating Toll-like receptors (TLRs), Nod-like receptors (NLRs), and RIG-I-like receptors (RLRs), collectively known as pattern recognition receptors (PRRs). It also protects cells from spontaneous ripoptosome formation—a multi-protein complex capable of killing cancer cells via caspase-dependent and caspase-independent mechanisms—by ubiquitinating RIPK1 and CASP8.
