Background

CHX10 functions as a transcriptional regulator by binding to the consensus sequence 5'-[TC]TAATT[AG][AG]-3' upstream of gene promoters. It plays a key role in the specification and morphogenesis of the sensory retina (By similarity) and may contribute to the specification of V2a interneurons during spinal cord development (By similarity). It mediates V2a interneuron differentiation by repressing motor neuron gene transcription through competitive binding to ISL1-LHX3 complex-activated response elements like VSX1. CHX10 acts as a positive transcriptional regulator of NXNL1, with significantly enhanced regulation in synergy with VSX1 (By similarity), while negatively regulating MITF (By similarity). It represses SAG transcription by competitively inhibiting ISL1-LHX3 response elements and binds to the photoreceptor conserved element-1 (PCE-1) in the SAG promoter as a transcriptional repressor (By similarity). CHX10 is involved in retinal ganglion cell (RGC) development, promoting Hedgehog signaling and retinal progenitor cell proliferation (By similarity). It contributes to inner nuclear layer cell development by promoting postnatal bipolar cell differentiation while inhibiting rod cell differentiation (By similarity). It may maintain neural retina identity during development by regulating canonical Wnt genes and CTNNB1 localization.
