Background

CHMP2B is a probable core component of the endosomal sorting required for transport complex III (ESCRT-III), which is involved in the formation of multivesicular bodies (MVBs) and the sorting of endosomal cargo proteins into MVBs. MVBs contain intraluminal vesicles (ILVs) generated by invagination and scission from the endosomal limiting membrane, primarily destined for lysosomal degradation of membrane proteins (e.g., stimulated growth factor receptors), lysosomal enzymes, and lipids. The MVB pathway requires sequential action of ESCRT-O, -I, -II, and -III complexes. ESCRT-III proteins typically dissociate from the invaginating membrane before ILV release. The ESCRT machinery also participates in topologically analogous membrane fission events, such as cytokinesis completion and budding of enveloped viruses (e.g., HIV-1 and other lentiviruses). ESCRT-III proteins are thought to facilitate vesicle extrusion and/or membrane fission, potentially in coordination with the AAA ATPase VPS4.
