CENPA is a histone H3-like nucleosomal protein specifically localized in centromeric nucleosomes. It replaces conventional H3 in the nucleosome core of centromeric chromatin, serving as an assembly site for the inner kinetochore. The presence of CENPA subtly alters nucleosome structure and DNA wrapping, resulting in less ordered and rigid protruding DNA ends compared to H3-containing nucleosomes. CENPA may act as an epigenetic mark propagating centromere identity through replication and cell division. It is essential for kinetochore protein recruitment/assembly, thereby regulating mitosis progression, chromosome segregation, and cytokinesis.
