Background

C/EBP α is a transcription factor that coordinates proliferation arrest and differentiation in myeloid progenitors, adipocytes, hepatocytes, and cells of the lung and placenta. It binds directly to the DNA sequence 5'-T[TG]NNGNAA[TG]-3', acting as an activator for distinct target genes. During early embryogenesis, it performs essential and redundant functions with CEBPB, being critical for the transition from common myeloid progenitors (CMP) to granulocyte/monocyte progenitors (GMP) and for proper liver and lung development (By similarity). It is also necessary for terminal adipocyte differentiation and postnatal maintenance of systemic energy homeostasis and lipid storage (By similarity). C/EBP α downregulates genes maintaining undifferentiated and proliferative states via E2F1 repression, crucial for adipocyte and granulocyte differentiation, while E2F1 reciprocally blocks adipocyte differentiation by binding promoters and inhibiting CEBPA's interaction with target gene promoters. Proliferation arrest further depends on functional binding to the SWI/SNF complex. In the liver, it regulates gluconeogenesis and lipogenesis through distinct mechanisms: cooperating with FOXO1 on IRE-controlled promoters (e.g., PCK1, G6PC1) for gluconeogenesis and synergizing with SREBF1 to activate lipogenic genes like ACAS2. In adipose tissue, it may act as a FOXO1 coactivator at the ADIPOQ promoter (By similarity). Additionally, C/EBP α can function as a dominant-negative, binding DNA with transactivation activity (albeit less efficiently than isoform 2) without inhibiting proliferation. It also directly enhances ribosomal DNA transcription by interacting with RNA polymerase I cofactors and inducing histone acetylation.

  • Western blot analysis of extracts from M-Liver (lane 1) and HepG2 (lane 2) using CEBP-α antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/2000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
