Background

CDK5RAP3 is a substrate adapter of E3 ligase complexes that mediates ufmylation, the covalent attachment of the ubiquitin-like modifier UFM1 to target proteins, and is involved in various processes such as ribosome recycling and reticulophagy (ER-phagy). As part of the UREL complex, it plays a key role in ribosome recycling by promoting mono-ufmylation of the RPL26/uL24 subunit of the 60S ribosome. This ufmylation occurs on free 60S ribosomes post-dissociation, weakening the junction between post-termination 60S subunits and SEC61 translocons to facilitate ribosomal subunit release and recycling from the endoplasmic reticulum membrane. The process can occur after normal translation termination or ribosome stalling during cotranslational translocation. Within the UREL complex, CDK5RAP3 acts as a substrate adapter that restricts UFL1 ligase activity to mono-ufmylate RPL26/uL24 at 'Lys-134'. The UREL complex also participates in reticulophagy under endoplasmic reticulum stress by ufmylating proteins like CYB5R3, promoting lysosomal degradation of ufmylated targets. Additionally, CDK5RAP3 regulates transcription by negatively modulating NF-kappa-B-mediated gene expression through RELA phosphorylation control. It governs mitotic G2/M transition and G2 DNA damage checkpoints and may induce MDM2-dependent p53/TP53 ubiquitination, stabilization, and nuclear activation via interactions with CDKN2A/ARF and MDM2, leading to G1 arrest and proliferation inhibition. It potentially contributes to nuclear envelope rupture during apoptosis and modulates MAPK14 activity by regulating its dephosphorylation by PPM1D/WIP1. Required for liver development (by similar). During microbial infection, it may be negatively regulated by hepatitis B virus large envelope protein mutant pre-s2 to promote mitotic entry.
