CD284 Antibody (YA5506) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to CD284.

Background

CD284 is a transmembrane receptor functioning as a pattern recognition receptor (PRR) that detects pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs) to initiate innate immune responses via downstream signaling pathways, including MYD88, TIRAP, and TRAF6, leading to NF-kappa-B activation, cytokine secretion, and inflammatory responses. At the plasma membrane, it cooperates with LY96 to mediate immune responses to bacterial lipopolysaccharide (LPS) and participates in LPS-independent inflammation triggered by free fatty acids (e.g., palmitate) and Ni2+. CD284 also facilitates TLR4 internalization via endocytosis, initiating MYD88-independent signaling through the TICAM1-TBK1-IRF3 axis to produce type I interferons. It further activates the NLRP3 inflammasome and establishes a positive feedback loop between autophagy and NF-kappa-B signaling. In complex with TLR6, CD284 promotes inflammation in monocytes/macrophages by interacting with CD86, and upon binding ligands like oxLDL or amyloid-beta 42, it triggers NF-kappa-B-dependent cytokine production (CXCL1, CXCL2, CCL9 via MYD88; CCL5 via TICAM1). In myeloid dendritic cells, CD284 mediates IRF7 activation by vesicular stomatitis virus glycoprotein G (but not LPS) to induce type I IFN production. Additionally, CD284 is essential for ZG16B/PAUF-induced migration of pancreatic cancer cells.
