CD223/LAG-3 Antibody (YA3842) is a Mouse-derived and non-conjugated IgG1 monoclonal antibody, targeting to CD223/LAG-3.

Background

CD223 (Lymphocyte activation gene 3 protein) is an inhibitory receptor primarily expressed on antigen-activated T-cells. It delivers inhibitory signals upon binding to ligands such as FGL1 (By similarity). FGL1 serves as a major ligand for LAG3 and mediates its T-cell inhibitory function (By similarity). Following TCR engagement, LAG3 associates with CD3-TCR in the immunological synapse, directly suppressing T-cell activation (By similarity). It may synergize with PDCD1/PD-1 to inhibit antigen-specific T-cell activation, potentially acting as a coreceptor for PDCD1/PD-1 (By similarity). LAG3 negatively regulates the proliferation, activation, effector function, and homeostasis of both CD8(+) and CD4(+) T-cells. Additionally, it mediates immune tolerance by being constitutively expressed on a subset of regulatory T-cells (Tregs) and contributing to their suppressive function (By similarity). It also acts as a negative regulator of plasmacytoid dendritic cell (pDC) activation (By similarity). While LAG3 binds MHC class II (MHC-II), its precise role remains unclear. It may function as a ligand for MHC-II on antigen-presenting cells (APCs), promoting their activation/maturation and driving Th1 immune responses.
