CD146 plays a role in cell adhesion, particularly in the cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. Its expression may facilitate interactions between melanoma cells and cellular components of the vascular system, thereby promoting hematogenous tumor spread. During embryonic development, CD146 could function as an adhesion molecule active in neural crest cells. Additionally, it acts as a surface receptor that induces tyrosine phosphorylation of FYN and PTK2/FAK1, along with a transient increase in intracellular calcium concentration.
