CD141 is an endothelial cell receptor that plays a critical role in regulating hemostasis, coagulation, fibrinolysis, inflammation, and angiogenesis. It acts as a cofactor for thrombin-mediated activation of protein C/PROC on vascular endothelial cells, initiating the activated protein C anticoagulant pathway. Additionally, CD141 accelerates the activation of plasma carboxypeptidase B2/CPB2, which inhibits fibrinolysis by removing C-terminal basic amino acids from substrates such as kinins or anaphylatoxins. It exerts protective effects by altering the cleavage specificity of protease-activated receptor 1/PAR1, thereby inhibiting endothelial cell permeability and inflammation (by similar). CD141 also suppresses inflammation independently of its anticoagulant cofactor activity by sequestering HMGB1, preventing its interaction with cellular receptors like RAGE.
