CD105 is a vascular endothelium glycoprotein that plays a crucial role in angiogenesis regulation. It is essential for the normal structure and integrity of adult vasculature and regulates the migration of vascular endothelial cells. CD105 may mediate endothelial cell shape changes in response to blood flow, facilitating vascular remodeling and the establishment of normal vascular morphology. Additionally, it may critically contribute to the binding of endothelial cells to integrins and/or other RGD receptors. Acting as a TGF-beta coreceptor, CD105 participates in the TGF-beta/BMP signaling cascade, ultimately activating SMAD transcription factors. It is required for GDF2/BMP9 signaling through SMAD1 in endothelial cells and modulates TGFB1 signaling via SMAD3. By similarity, CD105 is involved in extraembryonic angiogenesis and embryonic heart development.
