CBX4 is an E3 SUMO-protein ligase that catalyzes the sumoylation of target proteins by facilitating the transfer of SUMO from the E2 enzyme to the substrate. It is involved in the sumoylation of HNRNPK, a p53/TP53 transcriptional coactivator, thereby indirectly regulating p53/TP53 transcriptional activation and leading to p21/CDKN1A expression. Additionally, CBX4 monosumoylates ZNF131. CBX4 is a component of the Polycomb group (PcG) multiprotein PRC1-like complex, which is essential for maintaining the transcriptionally repressive state of numerous genes, including Hox genes, during development. The PcG PRC1 complex functions through chromatin remodeling and histone modifications, mediating monoubiquitination of histone H2A at 'Lys-119', resulting in heritable changes in chromatin expressibility. CBX4 also binds to histone H3 trimethylated at 'Lys-9' (H3K9me3) (by similar). It plays a role in germ layer lineage differentiation during embryonic development (by similar).

  Western blot analysis was performed on extracts from Raw264.7 (lane 1, 20 μg), C6 (lane 2, 20 μg), 293 (lane 3, 20 μg), PC-3 (lane 4, 20 μg), and Ramos (lane 5, 20 μg) using CBX4 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
