Background

CBFB, a core-binding factor, forms the heterodimeric complex, CBF, with RUNX family proteins, including RUNX1, RUNX2, and RUNX3. RUNX members exert transcriptional modulation on their target genes by recognizing the core consensus binding sequence 5'-TGTGGT-3', or exceptionally, 5'-TGCGGT-3', within regulatory regions through their runt domain. CBFB serves as a non-DNA-binding regulatory subunit that enhances the sequence-specific DNA-binding capacity of RUNX allosterically. These heterodimers bind to the core sites of various enhancers and promoters, such as murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL3, and GM-CSF promoters. CBF complexes play a role in the repression of the ZBTB7B transcription factor during cytotoxic (CD8+) T cell development, acting as a transcriptional silencer by binding to the RUNX-binding sequence within the ZBTB7B locus, thereby facilitating cytotoxic T cell differentiation. The interaction involves heterodimerization with RUNX1, RUNX2, and RUNX3, and CBFB also interacts with COPRS and forms a complex with PRMT5 and RUNX1.
