Caveolin-1 Antibody (YA549) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Caveolin-1.

Background

Caveolin-1 Protein plays a crucial role as a scaffolding protein within caveolar membranes, forming a stable heterooligomeric complex with CAV2, which targets lipid rafts and drives the formation of caveolae. This protein is involved in mediating the recruitment of CAVIN proteins (CAVIN1/2/3/4) to the caveolae, highlighting its intricate role in cellular signaling. Beyond membrane organization, Caveolin-1 directly interacts with G-protein alpha subunits, influencing their activity, and participates in the costimulatory signal necessary for T-cell receptor (TCR)-mediated T-cell activation. Additionally, its binding to DPP4 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner. Caveolin-1 further recruits CTNNB1 to caveolar membranes, potentially regulating CTNNB1-mediated signaling through the Wnt pathway. Notably, it negatively regulates TGFB1-mediated activation of SMAD2/3 by facilitating the internalization and subsequent degradation of TGFBR1 from membrane rafts. Through diverse interactions with various proteins such as BMX, BTK, CTNNB1, CDH1, GLIPR2, JUP, NOSTRIN, SNAP25, STX1A, SLC7A9, PACSIN2, EHD2, CAVIN3, and ABCG1, Caveolin-1 orchestrates a wide range of cellular functions, contributing to the dynamic regulation of membrane dynamics, signaling, and cholesterol homeostasis.

  • Western blot analysis of extracts from Hela (lane 1) and NIH3T3 (lane 2) using Caveolin-1 antibody. Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
