Background

cAMP is an antimicrobial protein integral to the innate immune system, binding bacterial lipopolysaccharides (LPS) and enhancing CXCL2 release via neutrophil N-formyl peptide receptors. Postsecretory processing generates cathelicidin antimicrobial peptides of varying lengths, serving as topical antimicrobial defense in skin sweat. The unprocessed precursor, cathelicidin antimicrobial peptide, inhibits Gram-negative E. coli and E. aerogenes with efficiency comparable to the mature peptide LL-37 in vitro. It induces membrane permeabilization by forming transmembrane pores, lyses E. coli, and exhibits broad-spectrum antimicrobial activity against Gram-negative (e.g., P. aeruginosa, S. typhimurium, P. syringae), Gram-positive (e.g., L. monocytogenes, S. epidermidis, S. pyogenes, S. aureus), and vancomycin-resistant enterococci. In low-salt media, it is effective against methicillin-resistant S. aureus, P. mirabilis, and C. albicans but inactive in 100 mM NaCl. It forms chiral supramolecular assemblies with P. aeruginosa quorum-sensing signal (PQS), potentially disrupting biofilm formation, and induces cytokine/chemokine production (e.g., TNF/TNFA, CSF2/GMCSF) in keratinocytes. It also displays hemolytic activity against red blood cells. Synergistic action with peptides KS-30 and KR-31 enhances bactericidal effects against S. aureus, E. coli, and C. albicans at lower concentrations while maintaining activity under high-salt conditions, though it cannot stimulate CXCL8/IL8 release from keratinocytes. Similar functions include: antimicrobial activity against E. coli and B. megaterium and hemolysis; synergy with KR-20 and KR-31; and weak antibacterial effects (MIC > 150 µM) or poor activity against E. coli K12.
