CASP8 is a thiol protease that serves as a molecular switch for apoptosis, necroptosis, and pyroptosis, playing a critical role in programmed cell death and preventing tissue damage during embryonic development and adulthood. As an initiator protease, it mediates the cleavage and activation of effector caspases (e.g., CASP3, CASP4, CASP6, CASP7, CASP9, and CASP10) to induce extrinsic apoptosis, contributing to FAS/CD95-mediated and TNFRSF1A-induced cell death. Binding to the adapter FADD recruits CASP8 to receptors FAS/TNFRSF6 or TNFRSF1A, forming the death-inducing signaling complex (DISC) for its activation. Additionally, CASP8 negatively regulates necroptosis by cleaving RIPK1 and initiates pyroptosis via cleavage of gasdermin-C and -D (GSDMC and GSDMD). In innate immunity, it cleaves and inactivates N4BP1 downstream of TLR3/TLR4 to promote cytokine production (by similar). It may also participate in Granzyme B (GZMB)-mediated cell death and cleaves PARP1/PARP2. Independent of protease activity, phosphorylated CASP8 (Tyr-380) promotes cell migration. Some isoforms lack catalytic sites and may inhibit caspase cascade activity or interfere with pro-apoptotic functions.
