Caspase-1 Antibody (YA5936) is a rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Caspase-1. It can be applicated for WB, IHC-P, ICC/IF, IP, ELISA assays, in the background of human, mouse, rat.

Caspase-1 is a thiol protease involved in various inflammatory processes by proteolytically cleaving other proteins. Its primary functions include: 1) Cleaving precursors of inflammatory cytokines interleukin-1 beta (IL1B) and interleukin 18 (IL18), as well as the pyroptosis inducer Gasdermin-D (GSDMD), into active mature peptides; 2) Serving as a key inflammatory response initiator in cell immunity - when activated through inflammasome complex formation, it initiates pro-inflammatory responses by cleaving IL1B and IL18; 3) Cleaving a tetrapeptide after an Asp residue at position P1; 4) Initiating pyroptosis (a programmed lytic cell death pathway) through GSDMD cleavage. Unlike IL1B cleavage, GSDMD recognition and cleavage doesn't strictly depend on the consensus cleavage site but relies on an exosite interface on CASP1 that binds the Gasdermin-D C-terminal part (GSDMD-CT). Additionally, Caspase-1 can: 1) Cleave and activate CASP7 during bacterial infection to promote plasma membrane repair; 2) Control antiviral immunity by cleaving and inactivating CGAS during DNA virus infections; 3) Cleave SPHK2 in apoptotic cells, which is released and remains enzymatically active extracellularly. Note that Caspase-1 is inactive in apoptosis. Similar functions include: GSDMD cleavage through exosite interface mechanism.
