Background

C9orf72 functions as a guanine-nucleotide releasing factor (GEF) for Rab GTPases by promoting the conversion of inactive RAB-GDP to active RAB-GTP. It acts as a GEF for RAB39A, facilitating HOPS-mediated autophagosome-lysosome membrane tethering and fusion during mammalian autophagy. As part of the C9orf72-SMCR8 complex, which exhibits GEF activity, it regulates autophagy. Within the C9orf72-SMCR8-WDR41 (CSW) complex, it serves as a GEF for RAB8A and RAB39B, promoting autophagosome maturation. The C9orf72-SMCR8 complex also displays GTPase activating protein (GAP) activity for RAB8A and RAB11A in vitro. This complex regulates autophagy initiation by interacting with the ULK1/ATG1 kinase complex and modulating its kinase activity. C9orf72 promotes autophagy initiation by regulating RAB1A-dependent trafficking of the ULK1/ATG1 complex to the phagophore, leading to autophagosome formation. It also modulates mTORC1 signaling by enhancing phosphorylation of mTORC1 substrates and plays a role in endosomal trafficking. by similar: May regulate phagosome-to-lysosome maturation; promotes lysosomal localization and degradation of CARM1, inhibiting starvation-induced lipid metabolism; regulates SMCR8 expression and synaptic localization in the forebrain, as well as protein levels of RAB3A and GRIA1/GLUR1 in the forebrain and hippocampus; restricts inflammation and autoimmunity in the hematopoietic system. Additionally, C9orf72 regulates actin dynamics in motor neurons by inhibiting ARF6 GTP-binding, leading to LIMK1/LIMK2 inactivation and CFL1/cofilin activation, thereby promoting axon extension and growth cone enlargement in spinal motor neurons. It is involved in stress granule assembly regulation under cellular stress but does not play a role in stress granule assembly regulation itself.
