c-Fgr Antibody (YA4792) is a mouse-derived and non-conjugated monoclonal antibody, targeting to c-Fgr. It can be applicated for WB, IHC-P, ICC/IF, ELISA assays, in the background of human, mouse.

c-Fgr is a non-receptor tyrosine-protein kinase that transmits signals from cell surface receptors lacking kinase activity, regulating immune responses including neutrophil, monocyte, macrophage, and mast cell functions, as well as cytoskeleton remodeling, phagocytosis, cell adhesion, and migration in response to extracellular stimuli. It promotes mast cell degranulation, inflammatory cytokine release, and IgE-mediated anaphylaxis, acting downstream of immunoglobulin Fc-binding receptors like MS4A2/FCER1B, FCGR2A, and/or FCGR2B. The kinase functions downstream of ITGB1 and ITGB2 to regulate actin cytoskeleton reorganization, cell spreading, and adhesion, exhibiting context-dependent activation or inhibition of cellular responses. While negatively regulating ITGB2 signaling, phagocytosis, and SYK activity in monocytes, it is essential for normal ITGB1/ITGB2 signaling and adhesion processes in neutrophils/macrophages. c-Fgr positively regulates cell migration via RAC1 activation and phosphorylates SYK (in vitro) to promote SYK-dependent AKT1 and MAP kinase signaling. In mast cells, it activates PLD2 through phosphorylation, generating lysophosphatidic acid and diacylglycerol, and promotes PIK3R1 activation. Additional substrates include FASLG (regulating its ubiquitination/internalization), ABL1, CBL, CTTN, PTK2/FAK1, PTK2B/PYK2, and VAV2. Notably, it only phosphorylates SYK-prephosphorylated HCLS1. When combined with CLNK, it negatively regulates NK cell-activating receptors and suppresses interferon-gamma production (By similarity).
