Background

Bok is an apoptosis regulator that functions through different apoptotic signaling pathways (PubMed:15102863, PubMed:20673843, PubMed:27076518). It acts as a pro-apoptotic protein that positively regulates the intrinsic apoptotic process in a BAX- and BAK1-dependent or -independent manner (PubMed:15102863, PubMed:27076518). In response to endoplasmic reticulum stress, it promotes mitochondrial apoptosis through downstream BAX/BAK1 activation and positive regulation of PERK-mediated unfolded protein response (By similarity). It activates apoptosis independently of heterodimerization with survival-promoting BCL2 and BCL2L1 by inducing mitochondrial outer membrane permeabilization in a BAX- and BAK1-independent manner, in response to inhibition of the ERAD-proteasome degradation system, leading to cytochrome c release (PubMed:27076518). During DNA damage response, it mediates the intrinsic apoptotic process in a TP53-dependent manner (PubMed:15102863). It plays a role in granulosa cell apoptosis through CASP3 activation (PubMed:20673843). Conversely, it functions as an anti-apoptotic protein during neuronal apoptosis by negatively regulating poly ADP-ribose polymerase-dependent cell death through modulation of neuronal calcium homeostasis and mitochondrial bioenergetics in response to NMDA excitation (By similarity). Beyond apoptosis, it may regulate trophoblast cell proliferation during early placental development by influencing G1/S transition via CCNE1 expression regulation (PubMed:19942931). It may also act as an autophagy inducer by disrupting the MCL1-BECN1 interaction (PubMed:24113155).

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
