Aurora B is a serine/threonine-protein kinase and a key component of the chromosomal passenger complex (CPC), which plays a central regulatory role in mitosis. The CPC ensures proper chromosome alignment and segregation at the centromere and is involved in chromatin-induced microtubule stabilization and spindle assembly. Aurora B mediates the bipolar attachment of spindle microtubules to kinetochores and regulates the onset of cytokinesis during mitosis. It also contributes to central/midzone spindle assembly and cleavage furrow formation, while functioning in the cytokinesis checkpoint by phosphorylating CHMP4C to delay VPS4-mediated abscission and prevent premature cytokinesis. Aurora B phosphorylates other CPC subunits, including BIRC5/survivin, CDCA8/borealin, and INCENP, enhancing its own kinase activity through positive feedback. Additionally, it targets multiple substrates involved in centromere function and mitosis, such as CENPA, DES/desmin, GPAF, KIF2C, NSUN2, RACGAP1, SEPTIN1, VIM/vimentin, HASPIN, and histone H3. In resting B- and T-lymphocytes, Aurora B phosphorylates H3S28ph at active promoters, inhibiting RNF2/RING1B-mediated H2A ubiquitination and enhancing USP16 deubiquitinase activity (By similarity). During mitosis, it phosphorylates the N-terminus of CGAS, blocking its liquid phase separation and activation to prevent autoimmunity. Aurora B also phosphorylates KRT5 (By similarity) and ATXN10, the latter promoting PLK1-mediated phosphorylation of ATXN10 and potentially regulating its proteasomal degradation.
