ATG3 Antibody (YA4523) is a mouse-derived and non-conjugated IgG1 antibody, targeting to ATG3. It can be applicated for WB, IHC-P, ICC/IF, FC, ELISA assays, in the background of human.

ATG3 is an E2 conjugating enzyme that catalyzes the covalent conjugation of the C-terminal Gly of ATG8-like proteins (such as GABARAP, GABARAPL1, GABARAPL2, or MAP1LC3A) to the amino group of phosphatidylethanolamine (PE)-containing lipids in the membrane, resulting in membrane-bound ATG8-like proteins—a key step in autophagosome formation. ATG3 cycles between binding to ATG7 (for loading ATG8-like proteins) and binding to the E3 enzyme (composed of ATG12, ATG5, and ATG16L1) to promote ATG8-like protein lipidation. Additionally, ATG3 acts as a membrane curvature sensor, facilitating LC3/GABARAP lipidation by sensing lipid-packing defects associated with high conical lipid ratios or sharp membrane curvature (by similar). It interacts with negatively charged membranes to enhance membrane tethering and LC3/GABARAP lipidation. ATG3 also functions as an autocatalytic E2-like enzyme, conjugating ATG12 to itself in an ATG7-dependent manner, forming a complex involved in mitochondrial homeostasis but not autophagy (by similar). The ATG12-ATG3 conjugate promotes late endosome-to-lysosome trafficking and basal autophagosome maturation via PDCD6IP interaction (by similar). This conjugate is also formed during vaccinia virus infection, disrupting cellular autophagy without affecting viral survival (by similar). Furthermore, ATG3 promotes primary ciliogenesis by removing OFD1 from centriolar satellites via autophagy (by similar).
