Atg16L1 Antibody (YA2187) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to Atg16L1.

Background

ATG16L1 plays an essential role in both canonical and non-canonical autophagy pathways. It interacts with ATG12-ATG5 to mediate the lipidation of ATG8 family proteins (MAP1LC3A/B/C, GABARAPL1/2, and GABARAP), facilitating autophagosomal membrane elongation. In canonical autophagy, it catalyzes phosphatidylethanolamine (PE) conjugation to ATG8, while in non-canonical autophagy, it likely conjugates phosphatidylserine (PS) to ATG8 during single-membrane autophagy at endolysosomal compartments (e.g., limiting IAV infection). Additionally, ATG16L1 regulates MAVS-dependent type I interferon (IFN-I) production, suppresses NOD1/NOD2-driven inflammatory responses, and promotes antibacterial autophagy with NOD1/NOD2. By similarity, it recruits LRRK2 to stressed lysosomes and restricts lethal IAV infection in epithelial cells via non-canonical autophagy.
