Atg12 Antibody (YA830)

Atg12 Antibody (YA830) is a Mouse-derived and non-conjugated IgE monoclonal antibody, targeting to Atg12.

Atg12 is a ubiquitin-like protein involved in autophagy vesicle formation. It conjugates with ATG5 via a ubiquitin-like system requiring ATG7 (E1-like activating enzyme) and ATG10 (E2-like conjugating enzyme). The ATG12-ATG5 conjugate functions as an E3-like enzyme essential for lipidation of ATG8 family proteins and their membrane association. As part of the ATG8 conjugation system (with ATG5 and ATG16L1), it recruits LRRK2 to stressed lysosomes and induces LRRK2 kinase activity (by similarity). In viral contexts, Atg12 may act as a proviral factor: when bound to ATG5, it suppresses type I IFN production to impair antiviral immunity (e.g., during VSV infection). It is also required for HCV RNA translation and replication initiation but dispensable for established infections.

  Immunohistochemical analysis of paraffin-embedded Rat heart tissue using Atg12 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80021, 1/200) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
