TMS1/ACS Antibody (YA5926) is a rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to ASC. It can be applicated for WB, ICC/IF, IP, ELISA assays, in the background of mouse, rat.

ASC is a pivotal mediator in apoptosis and inflammation. It promotes caspase-dependent apoptosis primarily via caspase-8 and caspase-9 activation (PubMed:11103777, PubMed:12646168), and participates in the mitochondrial apoptotic pathway by facilitating caspase-8-dependent BID cleavage, BAX translocation, and activation of caspases-9, -2, and -3 (PubMed:14730312, PubMed:16964285). In innate immunity, ASC serves as a core adaptor for multiple inflammasomes (e.g., NLRP1, NLRP3, AIM2), recruiting caspase-1 to process pro-inflammatory cytokines (PubMed:15030775, PubMed:17349957, PubMed:24630722). Its pyrin and CARD domains drive inflammasome assembly through homotypic interactions (PubMed:19234215, PubMed:24630722). ASC also engages in caspase-1-independent cell death (e.g., via caspase-8) (PubMed:19158675, PubMed:19158676) and modulates adaptive immunity by regulating dendritic cell maturation and DOCK2-mediated cytoskeletal reorganization (PubMed:22732093). It transcriptionally activates cytokines via AP-1/NF-κB pathways independently of inflammasomes (PubMed:12486103, PubMed:16585594) while suppressing NF-κB by inhibiting CHUK/IKBK kinase activity (PubMed:12486103). Recent studies suggest ASC cleaves CGAS to counteract DNA viral infection (PubMed:28314590). Notably, some evidence indicates ASC may inhibit inflammasome-mediated IL-1β maturation (by similar).
