Arp3 is the ATP-binding component of the Arp2/3 complex, a multiprotein complex that mediates actin polymerization upon stimulation by nucleation-promoting factor (NPF). The Arp2/3 complex facilitates the formation of branched actin networks in the cytoplasm, providing the force for cell motility. It appears to contact the pointed end of the daughter actin filament. In podocytes, Arp3 is required for lamellipodia formation downstream of AVIL and PLCE1 regulation. Beyond its cytoplasmic role, the Arp2/3 complex also promotes nuclear actin polymerization, regulating gene transcription and DNA damage repair. The complex enhances homologous recombination (HR) repair by driving nuclear actin polymerization, which mobilizes double-strand breaks (DSBs).
