Arg is a non-receptor tyrosine-protein kinase that shares overlapping roles with ABL1 in key processes related to cell growth and survival, including cytoskeleton remodeling in response to extracellular stimuli, cell motility, adhesion, and receptor endocytosis. It coordinates actin remodeling by tyrosine-phosphorylating proteins involved in cytoskeletal dynamics, such as MYH10 (movement), CTTN (signaling), and TUBA1/TUBB (microtubule subunits). Arg directly binds F-actin and regulates actin cytoskeletal structure via its F-actin-bundling activity. It modulates cell adhesion and motility by phosphorylating key regulators like CRK, CRKL, DOK1, or ARHGAP35. Adhesion-dependent phosphorylation of ARHGAP35 promotes its association with RASA1, recruiting ARHGAP35 to the cell periphery to inhibit RHO. Arg also phosphorylates receptor tyrosine kinases (e.g., PDGFRB) and substrates involved in endocytosis regulation (e.g., RIN1). In the brain, it may regulate neurotransmission by phosphorylating synaptic proteins. Additionally, Arg acts as a regulator of pathological signaling cascades during infection, where pathogens exploit its kinase activity to reorganize the host actin cytoskeleton for intracellular movement and cell exit. It self-regulates through autocatalysis and phosphorylation of its inhibitor, ABI1. By similarity, Arg positively regulates chemokine-mediated T-cell migration, polarization, and homing to lymph nodes and immune-challenged tissues, potentially via NEDD9/HEF1 and RAP1 activation.
