Background

ApoE is an apolipoprotein that primarily mediates lipoprotein-dependent lipid transport between organs through plasma and interstitial fluids. As a core component of plasma lipoproteins, it participates in their production, conversion, and clearance. This amphipathic protein interacts with both lipoprotein core lipids and the aqueous plasma environment, showing preferential binding to HDL while also associating with chylomicrons, chylomicron remnants, VLDL, and IDL. ApoE facilitates cellular uptake of lipoprotein particles by binding receptors such as LDLR, LRP1, LRP2, LRP8, and VLDLR. Its heparin-binding activity enables interaction with cell surface heparan-sulfate proteoglycans, promoting lipoprotein capture and receptor-mediated endocytosis. A key function involves hepatic clearance of chylomicrons, VLDL, and HDL, as well as VLDL biosynthesis and peripheral tissue uptake for triglyceride delivery. In reverse cholesterol transport, ApoE collaborates with ABCA1 in HDL biogenesis and mediates hepatocyte uptake of circulating HDL. Within the central nervous system, it regulates neuronal survival and sprouting. ApoE contributes to innate and adaptive immune responses (By similarity), including modulation of myeloid-derived suppressor cell survival, and binds immune receptor LILRB4. It may also influence transcription via a receptor-dependent, cholesterol-independent mechanism activating MAP3K12 and non-canonical MAPK signaling to enhance AP-1-mediated APP transcription. During microbial infection, ApoE interacts with HCV envelope glycoprotein E2 to facilitate viral attachment to HSPGs and other receptors (LDLr, VLDLr, SR-B1) on host cells, supporting HCV particle assembly, maturation, and infectivity.
