APE1 Antibody (YA2120) is a Rabbit-derived and non-conjugated IgG monoclonal antibody, targeting to APE1.

APE1 is a multifunctional protein that plays a central role in the cellular response to oxidative stress. Its major activities include DNA repair and redox regulation of transcriptional factors. Functioning as an apurinic/apyrimidinic (AP) endodeoxyribonuclease in the base excision repair (BER) pathway, APE1 initiates repair of AP sites in DNA by catalyzing hydrolytic incision of the phosphodiester backbone, generating single-strand breaks with 5'-deoxyribose phosphate and 3'-hydroxyl ends. It also cleaves AP sites in DNA/RNA hybrids, single-stranded DNA regions of R-loop structures, and single-stranded RNA molecules. At switch sites of immunoglobulin (Ig) constant regions, APE1 mediates Ig isotype class switch recombination (By similarity). APE1 exhibits 3'-5' exodeoxyribonuclease activity on mismatched deoxyribonucleotides during short-patch BER. Additionally, it acts as an endoribonuclease involved in single-stranded RNA metabolism regulation, such as controlling MYC mRNA turnover by cleaving between UA and CA dinucleotides in the MYC coding region determinant (CRD). In association with NMD1, APE1 participates in the rRNA quality control process during cell cycle progression.

  Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), 3T3 (lane 2, 15 μg), C6 (lane 3, 15 μg), K562 (lane 4, 15 μg), HCT116 (lane 5, 15 μg), and HuH-7 (lane 6, 15 μg) using APE1 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:30000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
